Results and Discussion ThroughouttheND5genesequencesused, neitherdeletionsnor insertionswere required formultiplealignmentandtheG十Ccontentswerenearly constant (21.0± 1.0%).Themaximumdifferencepercent inallsites,and thatatcodonthirdpositions intheND5genebetweenallthespeciesexaminedwerel4%and37%, respectively. 179 Zhi-HuiSしeta1 MultiplesubstitutionswerecorrectedbyK」MURA'sformula(KIMURA,1980).This,to- getherwithverylittledifferenceinG十CcontentofthegenethroughouttheCarabina- species,suggeststhelackofappreciableeffectsofbasecompositionbiasandmultiple substitutionson thephylogenetic trees.Partial discrepanciesbetween theUPGMA- andNJ-treeswere found forsomebranchingordersrepresentedby nodeswithlow bootstrapvalues.Theseinstabilitydidnotaffectthemainconclusionsinthispaper. F量i eonlineageswererecognizedinthetrees(Fig.1),i.e.,0homopterus,Car-abus (s.str),Alc11icata' bus,Apotomopterus (s. lat), Hemicarabus(s lat),Ast;1enocarabus, F)e,ntaca,・abus, Leptocalabus (s. lat), Mesoca,-abus, Autocarabus, 0,coca,-abus, Damaste''(s lat),Pt'ocrustes十Megodontus, C11rvsoca1-abus,andCllaetoca,-abus.The branchingorderofthese15groupswasnotconfidentlyresolveli,becausetheirbranch length(bootstrapvalueoflessthan70)isnotsignificantlydifferent fromzeroinmost cases. Ca,abus十Otic,noptet-us, Ast;1enocarabus十P,entaca,-abus, and Damaster十 Megodontus十Pl,oc1-ttstesmight respectively formonecluster.Evenif it isso, their branchingpointsaredeep. Theresultsinturnsuggest that the15groupsmentioned aboveemergedwithinashortperiod, i.e.,theyradiatedrapidlyintheinitialstageof theCarabinaevolution. Datingoftheradiationisdifficult.Takingthebasesubstitutionrateestimatedfor several arthropods'mitochondrial DNA(1.2X10-8/silentsite/year; BRowER, 1994), theCarabinaradiationmaybeestimatedtohavetakenplaceca llMyrago.This wouldbetheminimumestimationasdiscussedbySuetal.(1996a,b). Ifwesuppose thatthediversificationofDa,11aste,・(s.str)wouldhavestarteduponseparationofthe JapaneseArchipelago from theEurasianContinent, whichwasestimated tohaveoc- curredabout15Myrago,thentheradiationwouldhaveoccurredca 35Myrago(see SUetal.,1996b). SixFrench(European)genera,Atchica,abus(,feme,alls),Chaetocarabus(lntrica- tus),Atitoca'a' bus(atiratus),Mesoca'abus(p''oblematlcus), 0reocarablis(glab,atus), andChrvsoca'a' btis(atao' '1ltens)whichareabsent inJapan,donotshowcleardirect phylogenetic relations toany of theJapanese Carabina genera. Two Megodotltus- species,kolbel(Hokkaido,Japan)andvlolaceus(France) formacladeasexpected.C conc'/faro''issometimestreatedasasubspeciesofCa,-vensfs. Indee C conciliate・, (Hokkaido,Japan)isclosetoCan,ensls(France)ontheND5phylogenetictrees.Sup- pose that these twospeciesbranchedoff from their commonancestor somewherein theEurasianContinentandreachedFranceandJapan,respectively.Thebranchlength from thenode thatsupportsthese twospecies roughly representsthe relative time elapsedsincetheysplit. IfweassumetheCarabinaradiationtohaveoccurred35Myr ago(seeabove), then thebranch length of the two Ca,abus-speciescorresponds to 15Myr.Inotherwords,ca.15Myrwererequired for therespectivespeciestohavear- rivedatthepresentlocalitiesafter theirseparation. Thishasalsotoberelatedtothe timeofseparationoftheJapaneseArchipelagofromtheEurasianContinent,andmay strengthentheideathat theinitialCarabinaradiationisolder thangivenbyBRowERs' substitutionrate.
