However, during the past decade, spectacular advances have been made in the isolation and characterization of almost all of the other membrane systems of the cell, plastids, mitochondria, tonoplasts, nuclei, and other intracellular organelles. The availability of these new methods and the techniques involved are extremely important for advancing our understanding of plant cell physiology and biochemistry and for applying techniques of genetic engineering and molecular biology for the solution of basic and applied research problems. The diversity of approaches em- ployed by investigators interested in plant cell membranes indicated that this was an appropriate time to compile a methodological reference work for this area of plant biochemistry. Related methods and techniques which have been included in other volumes of this series have not been repeated unless recent developments represented significant improve- ments in the methods described previously.
