ExperimentalDesign:Dendriticcells(DCs)generatedfrompatientswithHER2-overexpressingbreast (n¼28)andpancreatic(n¼16)cancerweretransfectedwithDNAplasmidsthatexpresshumanHER2or heterologousratsequencesinseparateplasmidsoraschimericconstructsencodingrat/humanHER2fusion proteinsandusedtoactivateautologousTcells.ActivationwasevaluatedbyIFN-gELISPOTassay,perforin expression,andabilitytohaltHER2þtumorgrowthinvivo. Results:SpecificsustainedproliferationandIFN-gproductionbyCD4andCD8TcellsfromHER2-CP wasobservedafterstimulationwithautologousDCstransfectedwithchimericrat/humanHER2plasmids. Instead,Tcellsfromhealthydonors(n¼22)couldbeeasilystimulatedwithautologousDCstransfected withanyhuman,rat,orchimericrat/humanHER2plasmid.ChimericHER2-transfectedDCsfromHER2- CPwerealsoabletoinduceasustainedT-cellresponsethatsignificantlyhinderedtheinvivogrowthof þ HER2 tumors.Theefficacyofchimericplasmidsinovercomingtumor-inducedT-celldysfunctionrelieson theirabilitytocircumventsuppressoreffectsexertedbyregulatoryTcells(Treg)and/orinterleukin(IL)-10 andTGF-b1.
