Apoptosis
Apoptosis is the autonomous, orderly death of cells that are controlled by genes to maintain the homeostasis of internal

Caspase Family
Caspase is short for cysteine aspartic acid specific protease. It’s an important effector component of the apoptotic
mechanism and is involved in a variety of apoptosis related physiological and pathological processes. Caspase can be
divided into three groups: apoptotic initiators, apoptotic executioners and inflammatory mediators which form a cascade
amplification effect.

BCL-2 Family
There are many members in this family, such as Mcl-1, NR-B, A1, Bcl-w, Bcl-x, Bax, Bak, Bad, Bim, ect. All of them have
function of anti-apoptotic and apoptosis promotion. Most members have two structural homologous regions, which can
induce familial members to form dipolymers that is an important form for functional realization or functional regulation of
proteins.
When there is no death signal, Bcl-2 anti-apoptotic proteins are generally isolated as integral membrane proteins of the
organelle membrane, while pro-apoptotic proteins are distributed in the cytoplasm in an inactive form. When the cell is
stimulated with death signal, pro-apoptotic protein conformational change results from some proteases leading to not
only cell apoptosis which makes pro-apoptotic protein translocate to the membrane structure of organelles from cytosol,
also the reduction of anti-apoptotic protein inhibitory activity especially in mitochondrial membrane interacting with anti-
apoptosis protein.

Stem Cell
Stem cells are undifferentiated biological cells that can differentiate into specialized cells and can divide (through mitosis)
to produce more stem cells. They are found in multicellular organisms. In mammals, there are two broad types of stem
cells: embryonic stem cells, which are isolated from the inner cell mass of blastocysts, and adult stem cells that are found
in various tissues. In adult organisms, stem cells and progenitor cells act as a repair system for the body, replenishing
adult tissues. In a developing embryo, stem cells can differentiate into all the specialized cells—ectoderm, endoderm
and mesoderm (see induced pluripotent stem cells)—but also maintain the normal turnover of regenerative organs, such
as blood, skin or intestinal tissues.

CHOP, also known as GADD153 or DDIT3, is a highly conserved gene in both the structural
and regulatory regions to hamster gene. Imposed by unfolded and malfolded proteins,
CHOP is significantly induced by ER stress, deficiency of CHOP prevents cell from ER
stress. CHOP is considered a proapoptotic marker of ER stress dependent cell death. CHOP
acts as a dominant-negative inhibitor of the transcription factor C/EBP and LAP by forming
a heterodimer. It may play an important role in the malignant transformation of nevus to
melanoma.
